Insight on the regulatory elements responsible for dormancy process of Arabidopsis

چکیده
Dormancy is an important characteristics of seed to govern the seed germination at the right time of year. However, little information is available concerning the dormancy process at the molecular level. Clarifying molecular interactions in the cell necessitates detection and characterization of motifs in promoter sequences. To this end, the 2000bp upstream flanking regions of 22 genes corresponding to the dormancy process in Arabidopsis were analyzed to identify the upstream regulators of genes corresponding to the dormancy process. In this line, 42 promoter regions were obtained from Ensembl Plants. Using MEME, the first 20 significant motif sequences were identified as novel motifs with lengths ranging from 41 to 50bp. Motif 15, a binding site for C2H2, owned the most (700) occurrences. Moreover, motif 2 was found in 21 promoter sequences, which is the highest number in promoter sequences. The best match for this motif in Arabidopsis is BBRBPC_tnt.BPC1_col_a_m1, which is associated with the BPC1 transcription factor. The GOMo analysis predicted several biological functions such as response to chitin, regulation of transcription, protein amino acid phosphorylation, and response to abscisic acid stimulus for the 12 out of 20 discovered novel motifs. According to AME results, 215 consensus (known) motifs were enriched in 42 promoter sequences. The majority of consensus motifs were associated with AP2EREBP (37.2%), C2H2 (12%), C2C2dof (9.76%). In the current study, we identified several novel and consensus motifs and their corresponding transcription factors that possibly regulate the dormancy process and inhibit germination.کليدواژه ها
